「Seed-regeneration and Germplasm-processing Regulations」

Ⅰ General rules:

1. Short-term stored germplasm are regulated and conducted by the sixth part 「The utilization and management in short-term storage in the National Plant Genetic Resources Center」

2. The seeds need to posses sufficient quantity and high germination rate after selecting carefully before entering into storage. The seeds of low germination rate and

insufficient quantity are unable to store in storage until the original providers or the genetic center perform renewed regeneration plan to acquire the seeds of high germination rate and sufficient quantity and then put them into storage.

3. The seeds are divided into two parts after entering genetic center: one is for basic conservation in long-term storage. The other one is for active conservation in medium-term storage.

4. The purposes of basic conservation are for the perpetuation of seeds stocks. Basically, this kind of perpetual materials would not be used for exchanging and dispensing, but only for taking samples to perform the germination rate determination.

5. The purposes of active conservation (medium-term storage) come to the backup of basic conservation. The materials in medium-term storage are used for dispensing, exchanging, regenerating, estimating, proceeding germination rate determination periodically and renewing seeds. The consequences of germination rate determination would be the basis which whether renews seeds. The consequences of germination rate determination would be the basis which whether renews seeds and proceeds germination rate determinations of basic stored materials or not.

Ⅱ Quantity and quality

With regard to the quantity and quality of conserved germplasm materials which enter the storage, please follow the standard procedures as below.

1. Original seeds samples should follow the standard procedures as below.

(1) The seeds of cross-pollinated crop or multi-gene hybrids are 7,500 grains and upward.

(2) The seeds of pure line and autogamy line are 5,000 grains and upward.

(3) Different plants have different seed moisture content which had better keep between 6~12%.

(4) High pure seeds and the germination rate is 95% and upward (the lowest is 85%).

(5) The seeds are unable to reach the four points above which should be stored by using other ways, and be proceeded regeneration plan. Whereas, when the quantity and quality of seeds are suitable for active conservation (medium-term storage), the seeds could be proceeded active conservation temporarily and renewed regeneration plan.

(6) Because the germplasm regeneration possesses special limitations such as

regeneration specialty, environment and physiology, the germplasm regeneration does not need to follow the limitations above through the anction of genetic center.

2. Basic conserved (long-term conserved) standard:

(1) The seeds of cross-pollinated crop or multi-gene hybrids are from 3,000 to 4,000 grains.

(2) The seeds of pure line and autogamy line are from 1,700 to 3,000 grains.

(3) Different plants maintain different seed moisture content which had better keep between 6~12%.

(4) High pure seeds and the germination rate is 95% and upward (the lowest is 85%).

(5) The seeds are unable to reach the four points above which should be stored by using other ways, and be proceeded regeneration plan as well.

(6).Because the germplasm regeneration possesses special limitations such as regeneration specialty, environment and physiology, the germplasm regeneration does not need to follow the limitations above through the sanction of genetic center.

Ⅲ The conserved conditions

1. Basic conservation(long-term storage): The germplasm is conserved by using

aluminum pots to decompress and seal then be putting in the long-term storage with -12℃± 2℃ and RH30± 3%.

2. Active conservation (medium-term storage): The germplasm is conserved by using aluminum foil bags to seal and putting in the medium-term storage with 1℃± 2℃ and RH40± 3%.

Ⅳ Germination rate determination

1. The materials of active conservation (medium-term storage) should be periodically took samples to proceed the germination rate determination in accordance with different plants.

2. When the germination rate determination value reaches from 85% to 100%, the germination rate germination should be proceeded once every ten years except unique species.

3. When the germination rate determination value is lower than 85%, the seeds should be listed in the renewed regeneration plan and the germination rate determination should be proceeded every five years in order to estimate exact renewed time.

4. The germination rate determination of germplasm materials should detailed record every time in order to be the basis of building seed viability level data and drawing up determined plan and time.

Ⅴ Renewed regeneration of storing seeds 1. The standards of renewing regeneration:

The germplasm materials which stored in genetic center are mainly high quality seeds which should be selected carefully and the germination rate determination of high

If the seeds’ germination rate and quantity come to one of situations high quality below, the seed should be proceeded renewed regeneration.

(1) The germination rate:

The seeds with germination rate determination four times and upward should perform seed viability estimated mold to estimate the seeds of germination rate determination below 65% and the average of actual germination rage

determination below 65% and the average of actual germination rage determination below 65% in the last two times.

(2) Quantity:

Cross-pollinated crops or multi-gene hybrids seeds, for those which actual numbers of seeds multiply by the germination rage values are lower than 1,000 grains.

2. Operating direction

(1) Taiwan Agricultural Research Institute is responsible for drawing up, planning, coordinating, executing, dispensing germplasm materials and renewing

regeneration.

(2) The Council of Agriculture, Executive Yuan possesses agricultural research

institutes and domestic agricultural research organizations where are responsible for renewing regeneration of each kind of plant germination material according to the plans in Taiwan Agricultural Research Institute and techniques, experts, and geographic environments in every division.

Ⅵ 「The utilization and management in short-term storage in the National Plant Genetic Resources Center」regulation

1. Environmental conditions and capacity in short-term storage (1) Temperature 10℃± 2℃

(2) Humidity 40%RH± 3%RH

(3) Capacity: The length, width, and height of floorage in short-term storage are separately 11.8m, 7.4m, and 5.9m. There are fixed shelves in the short-term storage in order to place seeds neatly and manage seeds easily. There are Plastic frames in the fixed shelves. The plastic frame size is as below.

Large frames total 338 frames (Length*width*height: 62*43*15cm; which could contain 24 wide mouth bottles with 2.4 liters.)

Medium frames total 576 frames (Length*width*height: 62*43*24cm; which could contain 24 mouth bottles with 1.8 liters.)

Small frames total 504 frames (Length*width*height: 62*43*32cm; which could contain 24 wide mouth bottles with 1.2 liters.)

2. Usage

(1) The used objects:

The used objects are Taiwan Agricultural Research Institute, each transaction room in each substation, each agricultural organization and academic department.

(2)The stored objects:

The storage is for storing working materials such as breeding materials or other

related materials which carry specific gene, would be selected in the breeding plan or possess specific properties.

(3)Each division is able to request the materials in the short-term storage in genetic center, and only needs to fill in request form and material list which should be sent to genetic center.

3. Management

(1) The man needs to fill in stored form and material list when storing materials.

(2) The man should supply the request form and list of materials to genetic center, while the materials are stored in the storage.

According to stock operation, the manager in the genetic center inventories, weights, assigns numbers, stocks, prints the lists and then submits the lists to the division which provides the germplasm.

(3) The original divisions which request the germplasm form genetic center need to provide the request form and list with director’s autograph for the genetic center.

Then the genetic center would allot the materials and list to the divisions which request the germplasm.

(4) When the expiration date is due, the genetic center would notify the divisions which request the seeds to take back their materials. If the division doesn’t take back the materials, the genetic center would deal with the materials by itself.

(5) The staffs in the NPGRC would use the wide mouth bottles and plastic frames which are provided from the genetic center to put seeds for easy management and aesthetic.

(6) The materials in short-term storage do not conduct germination rate

determination and seed determination of moisture content, but for short-term storage only. Hence, it is suggested that the materials should possess high germination rate and be suitable moisture content when the materials are put into the short-term storage.

+ (Different plants maintain different moisture content. For example, rice moisture content should be lower than 13%.)

(7) The stored materials can not contain pests in order to avoid polluting other stored materials. If the materials carry with pests, the related divisions which provide the germplasm can inform the genetic center to fumigate the germplasm.

However, if the managers in the genetic center find out the germplasm which carry with pests, the managers can demand that the original provided divisions to fumigate the germplasm and then store into the storage.

4. The items what the genetic center provide

(1) Store in: Request forms, materials lists, packing containers (wide mouth bottles contain three sizes, large one comes to 2.4 liters, medium one comes to 1.8 liters and the small one comes to 1.2 litre.24 bottles for one frame and are placed on the shelf with the numbers), plastic frames and fumigation.

(2) Take out: Request forms and materials lists
