成果報告: NSC 94-2320-B-009-001 執行時間: 94/08/01 –95/07/31 主持人:楊昀良 國立交通大學生物科技學系 由於醫學的發展產生了各式各樣的醫療器材和藥物。這些新發展的普遍 應用,使免疫系統功能不健全的病患壽命延長,再加上抗生素的不適當使用, 使伺機型病原菌造成的感染病症大量地增加。其中尤以真菌感染的增加最為 顯著,在近年來已成為院內感染的主要病因之一。以美國為例,念珠菌 (Candida)已升為院內感染的第四常見致病菌。在台灣也不例外,念珠菌自 1981 至 2000 年間在北部某大醫院中造成院內感染的例子就增加了 16 倍,成 為最常見的院內感染的真菌致病原。因此,如何防治真菌感染已是一個刻不 容緩的課題。惟目前抗真菌藥物種類並不多,而且多半伴隨著危險的副作用; 尤有甚者,隨著大量使用抗真菌藥物,抗藥性菌株的出現也隨著增加。 產生真菌抗藥的主要機制之一是過度表現藥物幫浦CDR1,使真菌細胞可 將體內的藥物排出體外,減少藥物在體內的累積而產生抗藥性。為了瞭解
CDRI 表現的調控機制,我們建構了一個 lacZ 報告子(lacZ reporter)系統,
用來篩選白色念珠菌(Candida albicans)的 CDRI 的反式調控子
(trans-acting regulatory factors) 。我們將CDR1 的起動子(promoter)
和lacZ 基因結合,使得 lacZ 的表現受控於 CDR1 的起動子(CDR1p-lacZ)。
將此系統轉形至啤酒酵母菌後,再以基因遺傳(Gentics)表現之方法篩選白色 念珠菌的 gene library,進而找出 library 中能調控CDRI 表現的反式因子。
在篩選所得之基因中,被我們命名為REP1 的基因在過度表現(over
expression)時會提升CDR1p-lacZ 在啤酒酵母菌中的表現。進一步實驗結果
顯示,過度表現REP1 可改變啤酒酵母菌對抗真菌藥物的抗藥性。
本研究目的是了解REP1 在白色念珠菌中所扮演的角色。 經分析, REP1
和CaNTD80 有相似的 DNA binding domain,此 domain 與白色念珠菌產生抗
藥性有關。為了偵測REP1 是否與白色念珠菌抗藥性的產生及表現有關,我們
建構rep1/rep1 的突變株,以探討 REP1 在白色念珠菌抗藥性所扮演的角色。
此外,為進一步了解REP1 與 CaNDT80 的關係,我們亦建構 rep1/rep1 Candt80/Candt80 的雙突變株。以瞭解基因的功能。
