09/24* Culture E. coli DH5 (pCU1) in 3 ml LB (Amp), and E. coli DH5 (pBluescript SK+) in LB

(Amp) at 37oC overnight for plasmid DNA isolation.(前一天養菌,隔天收菌進行 Exp.3) (1) Plasmid (pCU1 and pBluescript SK+) isolation (Exp. 3),

(2) Restriction enzyme digestion (Exp. 5)

(隔天收並存於 4oC 供下一周使用,plasmid 存於-70oC 供下周使用)

(1) CIAP of pBluescript SK+ and fill-in DNA fragment from pCU1 (Exp. 8).

(2) Ligation of a DNA fragment from pCU1 to pBluescript SK+ vector with CIAP treated

(Exp. 9). (存於 4oC 供下一周使用). 6

(3) Competent cell preparation (Exp. 10). (存於-70oC 供下一周使用)

10/22(1) Transformation by heat shock and plate on LA (Amp). Incubate at 37oC overnight (Exp.11). (隔天觀察並存於 4oC)

12/10* Culture E. coli BL21 (DE3)(pET29a-A) in 3 ml LB (kanamycin), at 37oC overnight.

(前一天養菌,隔天收菌進行 Exp.18).

(1) Gene expression in E.coli and SDS polyacryamide gel electrophoresis of protein.

(Exp.18)(SDS-page 作好存於 4oC ,sample 煮沸後存在-70oC 供下一周電泳使用).

01/06 實驗期末考

基因工程實驗 (營養科學系)

(一) 上課時間、地點:

時間:每星期四上午8 :30 開始點名 (不補點名) 地點:LS301 教室

(二) 實驗規則:

1. 準時到堂上課,中途離開需先告知助教 2. 實驗桌上不可堆放書包衣物並保持清潔 3. 實驗室內禁止抽煙、吃東西、喝飲料 4. 學生於實驗期間,一律穿著實驗衣

5. 每組於實驗完畢需將實驗結果給助教簽退才可離開 6. 每組使用完畢之實驗器具交值日組簽收

7. 請珍惜實驗儀器及物品,如有損壞照價賠償,並扣期末分數五分。

(三) 作業:

1. 準備 A4 活頁紙

2. 上課前應預習實驗內容,以備小考

3. 實驗結果及紀錄於實驗結束時,交助教簽退 4. 結報由小助教統一收齊繳交至助教辦公室 123R

(遲交一日扣 10 分,遲交三日以上則該次作業不予計分)

5. 講義之後的問題及需於每次上課後各組自行討論解題,期中期末大小考由此出題,

並於適當的時間,請負責各部分的小組上台解題 6.作業內容:

本實驗課為二到三次實驗交一次結報,可不附預報,但須將各實驗之操作紀錄詳細 預報:

註1:點名不到者,超過 20 分鐘以曠課論;曠課三次即扣考

註2:請假注意事項:事假需事先請,病假可於當日請 (必須附上請假證明單) (五) 每週值日組負責工作:

1. 借音箱鑰匙及 LS301 鑰匙 (A 或 C 門) (系辦午休時間 12:00~PM1:00 勿打擾) 2. 抄黑板

3. 當日實驗組需最後離開
