一、 研究、教學、科技研發與管理工作全程經過概述。(由受聘人填寫)

前胸腺素 (Prothymosin α, 簡稱 ProT) 在細胞增生、免疫調節、乙醯化調節、以及調控氧化壓力 反應上扮演一定角色,然而前胸腺素的實際生理意義仍所知有限。我們先前研究發現前胸腺素過 量表現的基因轉殖小鼠會自發性產生肺氣腫,我們並且發現抑制小鼠肺部前胸腺素的表現能有效 減輕香菸造成的肺氣腫 (emphysema)。更重要的,我們發現在肺氣腫患者的肺組織切片中測定出 前胸腺素的過量表現且與肺氣腫的惡化情形呈現正相關,顯示肺氣腫致病過程中前胸腺素可能的 重要性。先前研究已指出,在肺氣腫動物模式中,乙型轉化生長因子(TGF–β)訊息傳遞的缺失可 能為其中一項主因,然而前胸線素如何調控乙型轉化生長因子訊息傳遞仍待釐清。因此,本計畫 主要目標釐清前胸腺素於乙型轉化生長因子訊息傳遞中的重要機轉以及探討前胸腺素過量於肺 氣腫及並惡化的發展。

具體工作項目如下:

1. 建立並繁殖前胸腺素過量表現基因轉殖小鼠

2. 建構前胸腺素基因干擾系統以抑制前胸腺素基因表現 3. 前胸腺素相關基因載體的製備

4. 前胸腺素蛋白的製備及純化

5. 前胸腺素與肺氣腫致病基因的相關訊息傳導細胞學實驗 6. 建立菸萃取物誘發肺氣腫的小鼠動物模式

7. 小鼠肺氣腫模式的治療

8. 臨床肺氣腫病人檢體的收集及分類整理

9. 分析臨床肺氣腫病人前胸腺素與其下游相關訊息傳導因子之實驗 10. 論文撰寫及投稿

我根據這些工作項目釐清前胸腺素在發炎及組織修復之間的相互關係,並探討前胸腺素在肺氣 腫形成過程中,前胸腺素對肺細胞的生理及肺氣腫病態生理學意義:

(1) 研究前胸腺素基因轉殖小鼠具有肺氣腫的表現型且分析其基質金屬蛋白酵素(MMP)及

組織抗基質金屬蛋白酵素(TIMP)之表現。

(2) 研究過量表現前胸腺素抑制 TGF-β 訊息傳導路徑。

(3) 研究前胸腺素調節發炎反應相關轉錄因子的活化而影響 TGF-β 訊息傳導路徑。

(4) 研究前胸腺素調節 TGF-β 訊息傳導路徑進而影響 MMP 及 TIMP 的表現。

(5) 進一步探討前胸腺素過量表現可能參與乙醯化調控發炎反應機制。

(6) 再進一步探討乙醯化調控與 TGF-β 訊息傳導路徑的關係。

此期間研究成果已發表至 Journal of Pathology 24th Oct 2015。

Roles of ProT in the development of emphysema. A proposed model depicting the HAT/HDAC imbalance caused by excessive ProT expression and the resulting imbalance between proteases and anti-proteases, leading to emphysema. Mechanistic connections among ProT overexpression, disabled TGF-β activation/ signaling, and proteases/anti-proteases imbalance. Imbalances in proteases and anti-proteases have been reported in COPD patients and various knockout mice exhibiting emphysema-like phenotypes. The impact of ProT on the HAT/HDAC imbalance has been described in our previous paper [1]. Fut8, α1,6-fucosyltransferase; Tg, transgenic.

研究進度已完成建立並繁殖前胸腺素過量表現基因轉殖小鼠,並將此小鼠進行菸萃取物引發 的小鼠肺氣腫模式。前胸線素過量表現基因轉殖小鼠以及肺氣腫患者的肺樣本中,抑制型Smad (I-Smad), Smad7 蛋白質增加,同時受體型 Smad (R-Smad), Smad2/3, 的磷酸化下降,以證明乙 型生 長 因子 -β的 訊 息 傳 導受到抑制,導致基質金屬蛋白酵素抑制物 (Tissue Inhibitor of Matrix Metalloproteinas, TIMP)表現下降,進而預期基質金屬蛋白酵素 (Matrix Metalloproteinase,

MMP) 的表現及活性增加,此為造成肺氣腫病徵的直接原因。根據這些完成之具體成果,我 們已整理這些成果發表,並預期更進一步提供臨床醫學研究及藥物開發於肺氣腫疾病之參考。

中心的計畫目標之ㄧ為更進一步探討人類疾病與細胞訊息傳遞相關機制。而受延攬人致力於 前胸腺素的研究,可有助於釐清抽菸與肺氣腫的部分相關機制,並提供新的研究方向。

(4)受延攬人於補助期間對貴單位或國內相關學術科技領域助益如何?

前胸腺素在細胞增生、免疫調節扮演重要角色,然而其中的機制仍須深入探討,而前胸腺素 真正在生物體疾病的生理意義尚待釐清。受延攬人參與前胸腺素與傳染性疾病及肺氣腫的研 究,有助於前胸腺素的基礎研究及臨床研究如癌症,多囊性腎腫,及肝硬化等疾病的關聯。

研究方面成果,已完成論文發表至 2015 年 Journal of Pathology, doi: 10.1002/path.4664. [Epub ahead of print]; 受延攬人同時有另一項針對癌症基因治療之研究:「Etoposide enhances antitumor efficacy of MDR1-driven oncolytic adenovirus through autoupregulation of the MDR1 promoter activity」已發表至 2015 年 Oncotarget, 6(35):38308-26.
