本實驗中,藉由CB-Span 85 結合逆流層析儀來萃取蛋白質混合 物。在樣品中加入卵白蛋白與溶菌酶混合液來進行萃取,發現雖然在 第 5 管時,溶菌酶會受到其它蛋白質靜電作用的影響而出現,在第 11管收集液中,仍然只有溶菌酶的存在,由於卵白蛋白不與 CB染料 分子作用,也就是對靜相沒有任何滯留力,在第 11 管的收集液,才 只有溶菌酶的存在。接下來使用溶菌酶與牛血清蛋白混和物當樣品,

CB 染料分子對其二者皆有親和力的影響,利用界面活性劑所形成水 核中心的大小來調控蛋白質進出反微胞系統,使溶菌酶被萃取,而牛 血清蛋白不被萃取進入反微胞系統中。拿溶菌酶、卵白蛋白與牛血清 蛋白之混合物來進行萃取,雖然溶菌酶會受到其他蛋白質的靜電作用 影響,使其回收率下降,但在CB-Span 85 所形成的反微胞系統下,

其反萃時收集液中只有溶菌酶的存在,這說明了CB-Span 85 只對溶 菌酶這種蛋白質有專一性的親和作用力。

在本實驗中,使用CB-Span 85結合逆流層析儀來萃取蛋白質混 合物這項技術,不僅製備容易而且價格低廉,是種應用性非常高的萃 取系統,接下來我們還可以使用其他種類之界面活性劑或加入其他的 萃取劑,來提升萃取效率,以及放大萃取容量,使得這項技術更具實 用性。我們預期具親和性的反微胞系統可以成為分離蛋白質的技術。
