Comparative potential therapeutic effect of sesame oil and peanut oil against acute monocrotaline (Crotalaria) poisoning in a rat model.

Many Crotalaria plant species contain hepatotoxic pyrrolizidine alkaloids (such as monocrotaline) that can cause acute and chronic poisoning in cattle and other animals. Peanut oil, atropine sulfate, and antidiarrheal agents are used to treat acute monocrotaline poisoning. The effect of sesame on acute monocrotaline poisoning has never been investigated. Monocrotaline significantly decreased (P < .05) serum amylase activity, but, over time, increased (P < .05) pancreatic and lung injury. AST and ALT activity and liver injury peaked at 24 hours. Sesame oil and peanut oil (P < .05) inhibited the changes in all tested parameters in acute monocrotaline poisoning. Although peanut oil inhibited acute monocrotaline poisoning, it induced steatosis, but sesame oil did not. We hypothesize that early pancreatic and lung injury and late liver injury contribute to acute monocrotaline poisoning and that sesame oil is more efficacious than peanut oil against acute monocrotaline poisoning in rats. However, additional studies are needed to confirm that these oils have the same effects in cattle and other animals.

Effect of sesamol and sesame oil on monocrotaline induced-sinusoidal obstruction syndrome in rats (Protective and therapeutic role).

Sinusoidal obstruction syndrome (SOS) occurs in patients undergoing hematopoietic cell transplantation and chemotherapy. The chemotherapeutic drugs oxaliplatin and cyclophosphamide cause SOS. Male Sprague-Dawley rats were gavaged with a single dose of sesame oil (0.5, 1, 2, or 4 mL/kg) One hour later, those rats were gavaged with monocrotaline (90 mg/kg) to induce SOS. and sesamol (5, 10, 20, and 40 mg/kg) was subcutaneously injected 24 h after monocrotaline treatment. Aspartate transaminase, alanine transaminase, laminin, collagen, myeloperoxidase, nitrate content, lipid peroxidation,

glutathione levels, matrix metalloproteinase (MMP)-9, and tissue inhibitor of matrix metalloproteinases (TIMP)-1 were assessed.

Effect of sesamol on isoproterenol-induced acute myocardial infarction in rats.

The protective role of sesamol and its possible action against isoproterenol-induced myocardial injury and infarction is unknown. We tested the hypothesis that sesamol's protection against myocardial infarction is associated with the inhibition of matrix metalloproteinase (MMP)-2 and MMP-9.Four groups of experimental rats were subcutaneously injected with sesamol (0, 1, 3, or 10 mg/kg) and then, 2 h later, intraperitoneally injected with isoproterenol (100 mg/kg 24 h apart on 2 consecutive days) to induce myocardial infarction. Blood pressure (BP), heart rate (HR), and electrocardiography (ECG) wave durations, serum creatine phosphokinase isoenzymes (CKMB), lactate dehydrogenase (LDH), myocardial histology, MMP-2, and MMP-9 were assessed 24 h after the last dose of isoproterenol was given.

Ostensibly ineffectual doses of cadmium and lipopolysaccharide causes liver damage in rats.

We found that co-exposing rats to ostensibly ineffectual doses of Cd and LPS significantly increased their AST, ALT, NO, peroxynitrite, and LPO levels but decreased their MT level. In addition, they underwent marked histopathological damage. The present study, however, clearly shows that even ostensibly ineffectual doses of Cd plus LPS injure rats’ livers. LPS initiated liver injury by increasing IL-1b levels and decreasing MT levels. Cd synergistically acted with LPS by increasing NO,

peroxynitrite, and LPO levels, but decreasing MT levels, which caused oxidative stress and liver damage.

In addition to proposed project, I was also involved in the following studies.

Effect of sesame oil on trinitrobenzene sulfonic acid induced Crohn’s disease in rats.

I have established Crohn’s disease model using trinitrobenzene sulfonic acid (TNBS) in rats and treated with sesame oil for 7 days. After 7-day treatment of sesame oil showed significant decrease in the weight/length, thickness, extension of lesion in colon, macroscopic, microscopic score and diahorrea. Preliminary data shows promising inhibitory role of sesame oil on Crohn’s disease.

Further studies are in progress.

Histopathological, histochemical and immunohistochemical staining

Apart from the other studies in progress, I have also done some considerable work in histochemical analysis in various disease and toxicology animal models. Extensive histopathological work was done in gentamicin induced renal damage and its recovery by sesame oil, which includes masson

trichrome, periodic acid schiffs, mast cell and AgNOR staining, in addition we have also done immunohistochemical staining for laminin in Crohn’s disease and the role of sesame oil.
