Chronic hepatitis B virus (HBV) infection can cause hepatocellular carcinoma (HCC). Among the mechanisms of HBV tumorigenesis, the viral oncoprotein HBx has been extensively studied. However, the exact mechanism for the HBx-induced tumorigenesis remains to be established. Recently, the molecular biology of liver regeneration (LR) has been used a potential model to study the mechanism of tumorigenesis. In this study, we therefore investigated the role of LR-related genes in HBx transgenic mice which developed HCC in 100% of transgenic mice at around 16 months. The dynamic expression of LR genes was analyzed at different stages (1M, 3M, 6M, 12M, non-tumor liver, and tumor) by Agilent cDNA array and analyzed by IPA software analyses. The candidate genes were then confirmed by real-time PCR. Consistent with array data, the hepatocyte-derived response genes, NF-B1 (Fig. 1) and c-Myc (Fig. 2), were significantly upregulated at the tumor part in the HBx transgenic livers.
Interestingly, SOCS family, which usually become activated in LR were divided into two opposing extremes in the HBx transgenic livers. SOCS4 and SOCS7 were significantly decreased at the tumor part, while SOCS1 and SOCS2 were increased at the tumor and non-tumor part of HBx transgenic livers, respectively (Fig. 3). Surprisingly, by Western blot assay, the expression of SOCS family was significantly decreased at the tumor part in HBx transgenic livers as compared to non-transgenic livers (Fig. 4). Base on these results, the activation of hepatocyte-derived response genes and the downregulation of SOCS family genes at the advanced stages may define the difference between LR and HBV tumorigenesis of hepatocytes. This dynamic expression profile of genes associated with LR in this model of HBx tumorigenesis should help to clarify the biologic regulation and difference in LR and HBV tumorigenesis.
