口腔癌為我國青壯年男性最容易罹患的癌症,且晚期存活率低於五成。因此,如何發展新穎天然藥物來當作癌症預防 或是治療藥物是目前重要的議題。厚朴酚(Magnolol)又稱為木蘭酚,為中國傳統藥物木蘭科植物厚朴(Magnolia officinalis)的主 要活性成份之一。Magnolol具有多種藥理功能,包含了抗菌、抗氧化、抗發炎、神經保護等功能。近年來許多文獻指出 Magnolol能夠抑制腦癌、乳癌、前列腺癌以及皮膚癌等癌細胞的生長,可經由活化或阻礙各種細胞內訊息傳遞,以達到抑制 細胞存活、增生、血管新生、侵襲及轉移並誘導癌細胞死亡。在這項研究中,我們研究了厚朴酚在人口腔癌細胞系中的抗 癌作用和機制。我們的結果表明,厚朴酚抑制口腔癌細胞HSC-3和SCC-9細胞生長並促進細胞凋亡。而由細胞凋亡蛋白酶晶 片與西方點墨法實驗顯示,厚朴酚增加了活化的caspase-3蛋白和血紅素加氧酶-1 (HO-1)的表達。此外,我們證明厚朴酚通過 調控MAPK pathway中的JNK1/2和p38路徑誘導口腔癌細胞系凋亡。綜合以上結論,厚朴酚可以做為口腔癌化學預防的潛在藥 物。
