3.4 討論
為了探討此技術的量測誤差[16],我們將誤差的討論就不同的樣品分為兩個 部分討論,其中誤差的主要來源有折射率的改變量有關△ 與入射角度的偏差n

將(3.3)式對透明物體折射率 n 做微分,我們可以得到

在n=1.33,反射光強度變化 I=0.001△ 時,可得相對應的入射光角度變化△θ=0.102°。在 n=1.463 的入射角度變化 θ=1.3△ 3°。但我們使用的旋轉台SIGMA SGSP-160YAW的精確 度可達0.0025°遠小於實驗可容許的角度誤差 θ=1.5△ 6°,故可以不考慮△ 的影響。所以θ 待測物體n=1.33 時本法的實驗解析度為 n=0.014△ 。
3.6 小結
本章討論利用反射光強度的方式,量測透明物質折射率分佈的方法及結果,為了驗 證其可行性,我們使用不同的待測物結構及待測物體,以及使用自行設計的驗架構配合 P 偏光以不同界面的布魯斯特角入射以量測二維折射率分佈,最後驗證了此方法的可行 性,也討論了此方法的誤差量。

4.2.3 四步驟移相法
[18-20]電光晶體調制器的快軸在x 軸上,並且在電光晶體調制器上施以瞘動電壓 ,如此出射光
Engineering Synthesis Design Inc.的IntelliWaveTM軟體,以及MathWork公司的Matlab軟體。
實驗時首先要調校實驗光路,雷射光通過電光晶體調制器後,經待測物反射通過檢偏 板並有干涉條紋的產生,這是由於電光晶體調制器是由兩塊晶體所構成,如下圖4.4 所示,
雷射光通過此電光晶體調制器後,p偏光與s偏光會有些微的夾角,最後經過檢偏板時產生 翦切干涉的現象,所以會有干涉條紋的產生。實驗時在移相之前,首先要先確定電光晶體 調制器移相時的驅動電壓,我們利用移相軟體IntelliWaveTM來替我們做校正的工作,如此
一來,可以避免因為初始相位 而造成移相時的誤差,所以經過校正後,我們可以得到實
驗時所需的驅動電壓。圖4.5a-d為電光調制晶體在移相 0、π/2、π、3π/2 時CCD抓取的 干涉圖形影像,接下來利用IntelliWave
Γ0
TM軟體計算後得到其相位差二維分佈圖,如圖4.6 所示,由此圖我們可初步判斷折射率的二維分佈,在圖中相位差為零的部分為篦麻油,這 是由於我們是利用空氣-篦麻油的布魯斯特角入射,並且由(4.10)式可以得知其干涉信號的 相位差為零。將圖4.6 所得到的相位差二維分佈圖代入(4.11)式計算,可以得到待測物折射 率二維分佈圖,如圖4.7 所示。

4.4 討論

4.5 小結
本章討論了量測透明物質折射率分佈的原理以及方法,為了驗證可行性,我們使用三 種油類的混合物當待測物,並且使用布魯斯特角的特性及移相干涉術實際量測相位的二維 分佈,最後驗證了此方法的可行性,也討論了此方法的解析度。

第五章 結論
本研究提出以布魯斯特角入射法,利用Fresnel’s equation,以強度及相位的 方式來量測透明物質的折射率分佈,並且使其成像,為了証明本法之可行性,我
們以嬰兒油、篦麻油及水和載玻片組成的樣品A 及樣品 B,其量測條件及結果
歸納如表5.1;因此可以驗證此方法並且達成
(1) 能分辨在平面上不同折射率的物質並且能抓取其分佈的區域。
(2) 利用布魯斯特角的特性,我們輕易的避開空氣-玻璃界面或空氣-水界面的 反射光,直接量測反射光強度,以得到待測透明物質的折射率分佈並成像。
(3) 相位測量法比強度測量法有較高的解析度。
且此技術有優點有:
(1) 操作容易。
(2) 利用現有相機取像軟體與程式軟體可以達成自動化的目標。
(3) 可以避免不必要的表面反射光。
(4) 可作待測物的長時間監測。
本研究可以應用於透明生物組織方面的研究,除了可以辨識生物組織的外型 之外,也可以利用監測其折射率的變化,研究當環境或是溶液成分有所改變時,
對生物組織所造成的影響。最後,未來針對此研究的努力方向有:
(1) 延續此技術,研究出布魯斯特角顯微鏡在量測透明生物體上的應用。
(2) 因為外差光源對於折射率量測的準確度更高,利用外差光源結合整面式相 位量測技術,是一個非常值得研究的方向。
