喜樹幼葉成分之研究
喜樹 (Camptotheca acuminata Decaisne) 為紫樹科 (Nyssaceae) 喜樹 屬植物,為原產於中國大陸的落葉性喬木,主要分布在四川、雲南跟 廣西等溫暖潮濕地區。喜樹裡最著名的二次代謝物是為喜樹鹼 (Campt othecin, CPT) ,在 1960 年代被發現具有良好的抗腫瘤活性。
本研究利用喜樹幼葉部以乙醇浸泡萃取,浸泡液經減壓濃縮後,濃縮 液經由水、乙酸乙酯以及正丁醇作分配萃取。取乙酸乙酯層及正丁醇 層部份分別利用矽膠管柱層析、全自動色層分析儀作初步分離,之後 利用薄層層析法、高效能液相層析法等方法,分離純化得到 41 個化 合物,其中包括九個 aliphatic 類、一個 monoterpenoid 類、一個 diterp enoid 類、二個 triterpenoid 類、一個 steroid 類、一個 pheophorbide 類
、六個 aromatic 類、七個 flavonoid 類、一個 norsesquiterpenoid 類、
一個 aliphatic glycoside 類、一個 secoiridoid glycoside 類、二個 quinoli ne 類、三個 quinic acid 類、四個 others 類,其中化合物 CA36 為新化 合物。
Studies on the constituents of the young leaves of Camptotheca acuminata .
Camptotheca acuminata Decaisne (Nyssaceae), a deciduous tree native to China, is widely distributed in relatively warm areas such as Sechwan, Yun nan, and Kwangsi. The secondary metabolites, Camptothecin (CPT) which was found to possess promising anti-tumor activity in the 1960s is isolated from C. acuminata.
In this study, the ethanol extract of young leaves of Camptotheca acuminat a Decaisne was concentrated to give a residue which was partition with wat er, ethyl acetate, and butanol. The ethyl acetate layer and butanol layer wer e separated by column chromatography on silica gel, automated flash chro matography, thin layer chromatography, and high performance liquid chro matography to afford forty-one compounds. Including nine aliphatic comp ounds, one monoterpenoid compound, one diterpenoid compound, two trit erpenoid compounds, one steroid compound, one pheophorbide compound, six aromatic compounds, seven flavonoid compounds, one norsesquiterpen oid compound, one aliphatic glycoside compound, one secoiridoid glycosi de compound, two quinoline compounds, three quinic acid compounds, an d four others compounds. Among them, CA36 is a new compound. .
