1. To clarify the role of autophagy in insulin secretion in β-cell
We used autophagy inducers and inhibitors to manipulate autophagy activity, and proved that autophagy positively regulates insulin secretion. Therefore, the lower autophagic activity of cell lines which are established from Min-6 cells by silencing Atg5 and Atg7 expression also confirmed that autophagy is essential mechanism for glucose stimulation insulin secretion.
2. To clarify the underlying mechanism of secretory autophagy regulation of insulin secretion
The autophagosomes were purified from Min-6 cells and identified engulfed protein by LC-MS/MS.
We demonstrated that autophagy engulfed whole insulin granules and identified Rab37 is a novel protein for autophagosome fusion with insulin granules. In addition, we created Rab37(-/-) cell line from Min-6 cells by CRISPR system and proved that Rab37 mediated secretory autophagy for autophagosome trafficking and autophagy related insulin secretion.
3. To clarify the role of autophagy in insulin secretion in vivo
We enhanced autophagy activity in B6 mice through fed autophagy inducer (Niclosamide) in diet.
In Niclosamide group, insulin level of blood was significantly upregulated and the the regulation ability of glucose homeostasis was improved. These data indicated that enhanced autophagy activity promotes insulin secretion in vivo.
4. To clarify the correlation between autophagy and insulin secretion in Type-II DM animal and clinical specimens
In two Type-II DM animal models including ob/ob mice and high fat diet induced mice, autophagy was activate and among colocalized with insulin granules inβ-cells. We further enhanced autophagy activity in B6 mice through fed autophagy inducer (Niclosamide) in high fat diet. Our data demonstrated that induction of autophagy improved hyperglycemia in type-II DM mice through enhancing insulin secretion.
