在本研究中,我們在飲用水中添加乙二醇,藉由增加攝取此種草酸 前驅物,我們已經成功地誘發大鼠腎臟內的草酸鈣結晶沉積,並進而形 成腎結石。而本實驗過程中並沒有大鼠因此死亡,表示使用此模式可以 有效地達成目的。
實驗後的生化尿液數據變化,可能與五苓散的作用有關,尚待進一 步證實。研究期間我們自行研發的圖片瀏覽程式,的確可以有效地簡化 統計分析工作。
灌食五苓散並飲用添加乙二醇水的大鼠,其體重增加情形與對照組 無異,推測五苓散可能是藉由減少草酸鈣結晶,因而減輕腎臟損傷,讓 大鼠能夠維持正常成長。
Alcohol dehydrogenase具有分解乙二醇的作用,本研究在正常大鼠 腎組織中,發現此酵素的蹤跡,意味著腎臟也具有分解乙二醇的功能。
我們在飲用乙二醇水的大鼠腎臟內,發現酵素AGT2的存在,代表著中 毒大鼠正積極加速glyoxylate此毒素的轉換清除工作。
經由腎切片結晶沉澱嚴重度評分結果,我們發現餵食五苓散的大鼠 腎結晶程度,明顯的低於只餵食安慰劑的大鼠。因此,我們證實灌食五 苓散可以顯著有效地減輕結晶的嚴重程度,五苓散在動物體內可以有效 的抑制草酸鈣腎結石。
而且,餵食五苓散可以有效的減少大鼠腎結石的發生率。同時,與 安慰劑組對比之下,餵食五苓散的兩組大鼠其罹患腎結石的相對危險性 也比較低。
因此我們可以歸納結論,五苓散的確是一個預防腎結石的有效方 劑。雖然,五苓散的抑石機轉尚未明瞭,推測尿液中的蛋白質應該扮演 著重要角色,期待日後能進一步深入研究。
