本論文選擇以系統生物學的工具探討神經保護中藥作用機轉實 質,主要是為了克服傳統研究方法局限在單點或單一機轉的限制,從一 個廣泛性的角度,來聯結一個藥物所具有的各種不同藥效之間的關係。

在銀杏萃取物的作用機轉研究中,探討當銀杏當作保健食品服用時,對 正常腦部的基因表現影響,其中 neuroactive ligand-receptor interaction 路 徑為銀杏在大腦中重要的調控路徑,在此路徑 amine 受體中的多巴胺受 體為主要調控對象,研究中並發現銀杏對腦部及腎臟的調控基因大不相 同。在丹皮酚的作用機轉研究中,探討神經細胞暴露在氧化壓力環境 時,丹皮酚所造成基因表現改變的整體影響,這些丹皮酚所調控的差異 表達基因勾繪出丹皮酚的抗氧化、抗發炎、及對神經相關基因的影響可 能是經由 NF-κB活性而得來,而與 NF-κB相關的 APP 抑制讓丹皮酚有

潛力成為一個預防Aβ產生的藥物。

研究的結果為銀杏及牡丹皮提供了作用機轉的基礎,丹皮酚能抑制 APP 的表現及銀杏對多巴胺受體的作用為牡丹皮及銀杏提供藥物新作 用的可能性,以供臨床上對銀杏在多巴胺缺乏疾病或是牡丹皮在氧化壓 力引起的相關神經學病變的應用。展望今後,除了對更多的神經保護藥 物進行實質作用探討外,中藥複方的神經保護作用也應有計畫的進行療 效分析,以期建立中醫藥神經保護作用更完備的臨床應用基礎。
