5-1 最佳電泳分析條件方面
(一) 利用MEKC/UV 電泳分離法,改變緩衝溶液中的成分百分比,
可以成功將九種色胺類物質標準品完全分離,九種色胺類物質包括:
α-Methyltryptamine (AMT)、N,N-Dibutyltryptamine (DBT)、
N,N-Diethyltryptamine (DET)、N,N-Diisopropyltryptamine (DiPT)、
N,N-Dimethyltryptamine (DMT)、N,N-Dipropyltryptamine (DPT)、
5-Methoxy-α-methyltryptamine (5-MeO-AMT)、
5-Methoxy-N,N-diisopropyltryptamine (5-MeO-DiPT)、
5-Methoxy-N,N-dimethyltryptamine (5-MeO-DMT)
(二)所使用分析方法的緩衝溶液(buffer)組成為:75 mM SDS、50 mM NaH2PO4溶液加入Water-Methanol-Acetonitrile (65:30:5,v/v ) 後,並且以磷酸 調整pH至2.1,然後對標準品進行分離,發現微胞電動層析法 ( MEKC ) 的 最低偵測極限介於DBT ( 0.23 ppm )與5-MeO-DiPT ( 0.80 ppm )之間,而毛細 管電泳掃集法( sweeping- MEKC ) 的最低偵測極限介於AMT ( 1.10 ppb )與 5-MeO-DiPT ( 4.60 ppb )之間。
5-2 應用與展望
本實驗所研發之分析方法,具有高靈敏度,快速且經濟,目前以改 變水溶液中的有機相及十二烷基磺酸鈉 ( SDS ) 濃度來進行九種色胺類 物質標準品的分離,可針對水溶液中或有機相中的上述九種色胺類物質
( CSEI-sweep-MEKC )進行分離,並期望進一步降低其偵測極限,期盼 建立一套快速、有效又具有可信度的分析方法。
