附錄一 實驗儀器

(Olympus, IX71, IX73),連接彩色攝影機後搭配相位差濾鏡便可拍攝較為立體之 生物樣本照片;連接汞燈並搭配螢光濾鏡後便可分別拍攝不同顏色之螢光照片,

最後再利用電腦軟體疊圖。

即時細胞培養觀察系統與細胞培養箱

本實驗使用之細胞培養箱包含裝設有即時攝影系統的培養箱(Cultured cell monitored system, ASTEC)以及一般培養箱(MCO-19AIC, Panasonic),用來維持體 外細胞培養所需之環境以及拍攝細胞生長狀態的照片。

無菌操作臺

本實驗使用無菌操作臺(4BH-24, HIGH TEN Scientific Corp.)是為了建立一無 菌環境,並在此環境下操作細胞相關實驗。

DAQ 介面卡

為了使用Labview (National Instrument)電腦軟體來控制電極的電壓施加與否,

使用DAQ 介面卡(USB-6509, National Instrument)連至電路板來進行控制。

繼電器電路板

電訊號經電壓放大器放大後傳入繼電器電路板,本實驗使用之電路板含有 48 個繼電器(LU-5, RAYEX)以控制各個腳位的開啟或關閉,而繼電器的開關則由 電腦軟體Labview (National Instrument)連接 DAQ 介面卡與電路板來控制。此電 路板之輸出電壓會聯接一具有48 個腳位的排線,最後接至加裝帶狀電纜連接器 (CC-125FRRC, Yokowo)以利輸入訊號至電控微流體平臺上。

點光源曝光機

本研究使用此UV 曝光設備(OmniCure Series 2000, Lumen Dynamics)用來交 聯水膠預聚物溶液,其輸出之光線波長介於320 nm-500 nm,可自行設定曝光時 間,照度則介於 0.2-40 W/cm2,可藉由原廠的 UV 照度計(OmniCure radiometer, R2000-2025, Lumen Dynamics)進行量測,該照度計之準確度為 5%,量測波長範 圍介於250 nm-1000 nm,量測功率介於 1-12000 mW。

動態 ELISA 讀盤機

本研究使用動態ELISA 讀盤機(Vmax, Molecular Devices)檢測實驗中細胞分 泌物經過ELISA 方法成色後的吸光值,並依照比色法計算出該分泌物之含量。
