1. 本研究顯示,在所評估之濃度 23 μg/mL EMA 及 PMA,於螢光顯微鏡下觀察,
EMA 或 PMA 皆可進入受熱細菌的細胞膜內,但 EMA 也會進入完整細胞膜 內,而 PMA 較無此情況。
2. 本研究顯示,在所評估之各濃度 EMA 及 PMA 中, 1.5 μg/mL PMA 可有效 抑制受熱組細菌於 qPCR 放大,且在此濃度下,不會對活性細菌造成 qPCR 定量濃度之干擾。因此以1.5 μg/mL PMA 濃度做為結合 qPCR 定量空氣中活 性細菌之最佳條件。
3. 進一步以最佳核酸染劑條件 1.5 μg/mL PMA 評估細菌濃度定量範圍,結果顯 示,最佳線性範圍介於 1×104 ~ 1×1010 cfu/mL。
4. 將開發之方法進行職業場所空氣採樣之樣本分析,量測場所包括畜牧場、家 禽舍、木材行、水稻田、蔬菜田及醫療院所。結果顯示所開發之 PMA/qPCR 方法確實可作為定量空氣中活性細菌之分析方法。
