第五章、結論

大白鼠在經水迷津試驗後會顯著降低海馬迴CA1 區域內 STAT1 mRNA 與蛋白質的表現量,但此現象並不發生於杏仁核及紋狀體區域,此結果顯 示 STAT1 可能參與海馬迴 CA1 區域所調控的空間記憶學習。論文利用 STAT1 siRNA 轉染至海馬迴 CA1 區域抑制 STAT1 的表現量,進而促進大 白鼠在水迷津試驗的空間學習能力。然而,大白鼠轉染STAT2 siRNA 則不 會影響大白鼠在水迷津試驗的空間學習能力,證實 STAT1 在海馬迴 CA1 區域扮演著抑制空間記憶形成的角色。本實驗室於2009 發現 laminn β1 的 表現量下降會促進大白鼠在水迷津試驗的空間學習能力 (unpublished observation, 附錄二),而本論文在 laminin β1 報導基因實驗發現,STAT1 siRNA 會降低 laminin β1 1.3kb 啟動子的轉錄活性;反之,STAT1-Flag 則 會增加 laminin β1 1.3kb 啟動子的轉錄活性,顯示 STAT1 調控 laminin β1 的轉錄能力。此外,本論文於動物實驗中發現,轉染STAT1 siRNA 至大白 鼠海馬迴CA1 區域會降低 STAT1 與 GAS 序列的結合能力及 laminin β1 的 表現量;反之,大白鼠轉染野生型 STAT1 質體則會增加海馬迴 CA1 區域 內 STAT1 與 GAS 序列的結合能力及 laminin β1 的表現量,顯示 STAT1 正 向調控 laminin β1 的表現。綜合上述的研究結果,顯示 STAT1 在海馬迴 CA1 區域抑制大白鼠空間記憶的學習能力,此現象可能與 STAT1 正向調控 laminin β1 的表現有關聯。
