營養生化指標是評估營養狀況的重要方法。我國過去進行的國民營養健康狀況變遷調查所分析的營養生化指標有限,且仍 有部分人群的營養狀況需持續追蹤。本研究擬使用國民營養健康狀況變遷調查所收集的血液樣本,分析血中視網醇
(retinol)、α-生育醇(α-tocopherol)、β-胡蘿蔔素(β-carotene)、運鐵容量(total iron binding capacity, TIBC)及磷酸比哆 醛(pyridoxal-5’-phosphate, PLP),評估國人維生素 A、維生素 E、β-胡蘿蔔素、鐵及維生素 B6 的營養狀況。鑒於未來國民營 養健康狀況變遷調查需持續進行大規模樣本的分析,發展快速、靈敏度佳且樣本需求量少的分析方法有其必要性。在脂溶性 營養生化指標方面,本研究擬以高效能液相層析儀(HPLC)同時分析血中維生素 A、維生素 E 及 β-胡蘿蔔素濃度。在鐵的
營養生化指標方面,本研究擬以血清TIBC 作為鐵的營養狀況指標,將採用血液生化自動分析儀分析。在維生素 B6 的營養
生化指標方面,本研究將以改良傳統之HPLC 法偵測血中 PLP,已證實其操作時間可由 1 小時縮短至 10 分鐘內,亦可將血 漿樣品需求量由100-200 μL 減少至 50 μL。本研究有助於了解國人維生素 A、維生素 E、β-胡蘿蔔素、鐵及維生素 B6 的營養狀 況,並建立國民營養狀況標準生化分析方法。
• 英文摘要 Analysis of nutritionally biochemical markers is an important method for assessment of nutritional status. The previous Nutrition and Health Surveys in Taiwan (NAHSIT) have analyzed limited nutritionally biochemical markers and suggested that it is necessary to monitor the nutritional status of certain people. The purpose of this study is to assess the vitamin A, vitamin E, β-carotene, iron, and
vitamin B6 status of people in NAHSIT by measuring their blood retinol, α-tocopherol, β-carotene, total iron binding capacity (TIBC), and pyridoxal-5’-phosphate (PLP) levels. It is critical to develop rapid and sensitive methods for future NAHSIT. For the analysis of fat-soluble markers, a simultaneous analysis of retinol, α-tocopherol, and β-carotene levels will be conducted using HPLC. For the analysis of blood total iron, TIBC will be measured using an autoanalyzer. For the analysis of blood PLP, a modified HPLC method will be conducted. We have approved that it is a time-sparing (10 min vs. 1 h) and sample-sparing (50 μL vs. 100-200 μL) method. This study will contribute to understand the vitamin A, vitamin E, β-carotene, iron, and vitamin B6 status of people in Taiwan and to establish standards for biochemical assessment of nutritional status.
